ABSTRACT

Gentianae Radix et Rhizoma has a long history of medication, and was recorded in Shennong Bencaojing. Through the study of herbal medicine of Gentianae Radix et Rhizoma,we reviewed and analyzed previous herbal records, studied its basic, origin and processing methods, and provided the basis for further development and utilization of Gentianae Radix et Rhizoma. Through the analysis of ancient herbal researches and drawings,we knew that the ancient resources of this medicine were basically consistent with Gentiana scabra and G. manshurica. The Latin name of the original plant of gentian medicinal herbs set forth in the modern literature was analyzed for the first time. The commodities of this medicine were divided into Guanlongdan and Dianlongdan. Guanlongdan was produced in Northeast China, and consisted of G. scabra,G. manshurica, and G. triflora. Dianlongdan was G. rigescens and grew in Yunnan province. Due to various reasons, such as historical changes, genuine producing areas of medicinal materials gradually changed from Jiangsu and Zhejiang to Northeast China. The processing methods of Gentianae Radix et Rhizoma included preparing with wine, preparing with licorice juice and preparing with ginger juice. The purpose of these processing methods is to suppress its nature of bitter and cold. Among those processing methods, preparing with wine has the longest history, and can promote the dissolution of the active ingredients of gentian, thus is a better method of processing. This study provides the basis for radical reform, resource development, correct use, and determination of genuine producing area and processing methods for Gentianae Radix et Rhizoma.

ABSTRACT

A rapid and accurate method was established by psbA-trnH sequence to distinguish Dioscorea nipponica from other species belonging to the same genus. In this study, we collected 144 samples of D. nipponica from 17 different producing areas in China, the psbA-trnH sequenced of 23 nucleotide sequences were downloaded from GenBank, the sequences from Dioscorea genus. DNAMAN 8.0 software was used to show splicing, MEGA 6.0 software was applied for sequence analysis and comparison. The results showed that the genetic relationship between D. nipponica and D. subcalva was the closest, and the genetic relationship with D. zingiberensis was the furthest. It is indicated that the psbA-trnH sequence as a DNA barcode can effectively distinguish between D. nipponica and other plants in the genus of Dioscorea.

ABSTRACT

Mitogen-activated protein kinases (MAPKs) signal is one of the important ways in eukaryotic cell, which adjusts and controls the structure and function of the cell. MAPKs in eukaryotes include p38, ERK, JNK and ERK5, etc. With the deepening research,we found that the activation of p38, ERK, JNK signal pathways were closely related with osteoarthritis (OA) cartilage injury. MAPKs are the key signaling systems involved in the production of matrix metalloproteinases and the regulation of cartilage cell proliferation, apoptosis and differentiation. Expecially the matrix metalloproteinases can accelerate the degradation of articular cartilage. So it has been the new spot in pathogenesis of osteoarthritis study.

ABSTRACT

Objective: To analyze the genetic diversity of Geranium wilfordii from different habitats at DNA molecular level and to establish the ISSR-PCR system of G. wilfordii. Methods: Using orthogonal design test combined with the single factor gradient optimization method, the geranium ISSR-PCR system of G. wilfordii was determined. G. wilfordii from 20 germplasmic resources was amplified and analyzed by ISSR-PCR. After translating the amplification results into "0" and "1" matrix, the genetic distances were calculated by Popgene32 and Ntsyspc-2.1 software and the systematic diagram of genetic relationship was clustered by UPGMA method. Results: A total of 207 bands were detected using 15 primers, among which 197 (94.6%) were polymorphic bands. The genetic distances were changed from 0.2018 to 0.9394. The genetic relationship of G. wilfordii was more complex. The 20 samples were divided into two kinds, and the correlation between the most germplasm and its geographic origin had a certain relationship. Conclusion: The significant polymorphism and genetic diversity are observed among G. wilfordii germplasm resources which could provide a wealth of genetic basis for the germplasm high quality.

ABSTRACT

Osteoarthritis is a common joint disease, which seriously affects the patient's health and quality of life. It results in substantial social and economic costs. Etiology and pathogenesis of OA is still not completely clear, but people paid more attention on Cytokines, especially IL-1, which is considered as core factor in the development of OA. In recent years, many clinical trials considered IL-1 as a target treatment for OA. It provided a new treatment method. This article is to overview the mechanism of IL-1 in OA cartilage damage.
